EpiSCs were plated in their maintenance Fgf2 + ActivinA + XAV (FAX) medium. iEsrrb-driven reprogramming was induced by switching to 2i+doxycycline (dox) the next day. After 4 days of reprogramming, Dox was withdrawn and blasticidin (bsd) was added to select for naïve colonies with an active Rex1 promoter.
