(A) Schematic overview of Heparinase I/III digestion of HS chains located on HSPGs on the cell surface. Cells were pre-treated for 2 h at 37°C with 1 Unit Heparinase I/III. Subsequently, cells were washed and infected with VSV WT or VSV-GP variants (MOI 1) for 1 h at 37°C in complete growth medium. (B) Flow cytometry analysis of cell surface HS of Heparinase I/III treated and untreated 293T WT cells. Infection of Heparinase I/III treated (C) 293T WT and (D) ΔDAG1 cells with VSV WT and high or low affinity VSV-GP variants. Infection was quantified by flow cytometry measuring eGFP signal 15 h p.i. Shown are the means ± SD of three replicates.
