Cells were grown in YEPD medium at 30°C (acc1cs) or 23°C (acc1ts) to an OD600 of 0.3. The cultures were split; one fraction was maintained at the permissive temperature, and the other was shifted to the nonpermissive conditions for 3 or 4 h, respectively. Cells were harvested, and cytosol was prepared as described in Materials and Methods. Enzyme activities were determined as described elsewhere (25, 26). The standard deviation of four experiments was below 10%. ND, not detectable.
