Effects of H. zeylanica-E2 on PGE₂ expression mediated through COX-2 expression. AGS cells were treated with 10 μg/mL H. zeylanica-E2, and BGC823 cells were treated with 5 μg/mL H. zeylanica-E2 for 0, 24, or 48 h. The COX-2 mRNA transcript and protein expression levels were determined using (A) quantitative reverse transcription polymerase chain reaction (qRT-PCR) and (B) Western blotting. GAPDH was used as an internal control. (C) The PEG₂ protein level was measured using an enzyme-linked immunosorbent assay (ELISA). (D) AGS and BGC823 cells were treated without or with H. zeylanica-E2 or 5 μM CLC for 24 h. The conditioned medium was collected and the PEG₂ level was measured by ELISA.
