BMH-21 disrupts the retention of EGFP-TOP2B in the nucleolus. (a, b) Rapid dispersion of nucleolar EGFP-TOP2B by BMH-21. Experiments were performed on EGFP-TOP2B as described in Fig. 6b and c. (c) A control experiment using DMSO, instead of BMH-21. (d, e) Quantification of the nucleolus-to-nucleoplasm ratios of EGFP-TOP2B. Experiments were performed using 1 µM (d) or 2 µM (e) BMH-21. Average values and SD were calculated (n = 10). (f, g) BMH-21 treatment does not affect the nucleolar localization of nucleolin and fibrillarin in ATP-depleted cells. HeLa cells expressing either EGFP-nucleolin (f) or EGFP-fibrillarin (g) were incubated with 25 mM 2DG/10 µM CCCP for 20 min and subsequently treated with 2 µM BMH-21 for 5 min in the presence of 2DG/CCCP.
