PLIN2/LDs regulate GSIS in EndoβCH2-Cre cells. A: GSIS was blocked in PLIN2KD cells compared with Sham, whereas this was enhanced in PLIN2OE. Levels were first normalized to DNA content and then to Sham at 2.5 mmol/L glucose. Preproinsulin mRNA (B) trended higher in both PLIN2OE and PLIN2KD cells, although insulin protein levels (C) were only elevated in PLIN2KD. Normalization was to 18S (B) or DNA content (C) before Sham. D: The heat map shows that insulin secretion–related gene expression was generally decreased in PLIN2KD cells, while it was elevated in PLIN2OE cells. The color keys represent the row-wise Z score.
