KSM-66 inhibits protein glutathionylation. (A) Glutathionylation levels were detected by using Western blot. Cells were pre-treated with or without 0.5 mg/ml KSM-66 for 24 h followed by 100 μM 6-OHDA for 15 min. Total proteins were separated on 12% SDS-gel under non-reducing conditions (no DTT) for Western blot analysis using anti-glutathione mouse monoclonal antibody. (B) Relative major glutathionylated band (GSSG) was normalized with GAPDH loading. Data are mean ± SEM of three independent experiments. Data were analyzed by One-way ANOVA with Bonferroni's multiple comparison test. No significant differences (P > 0.05) in expression profiles were observed for control, 6-OHDA and KSM treatments.
