Some HRAS-overexpressing HNSCC PDX models are sensitive to tipifarnib. BALB/c nu/nu or SCID mice were inoculated subcutaneously with 2–3 mm tumor fragments, the PDX were allowed to establish to 250–350 mm3, the animals were randomized into groups of three and treated orally BID with vehicle or tipifarnib (60 mg/kg for SCID, 80 mg/kg for nu/nu) for 25–30 days. Tumor volumes were measured twice weekly in two dimensions using a caliper, and the volume was expressed in mm3 using the formula: V = (L × W × W)/2, where V is tumor volume, L is tumor length (the longest tumor dimension), and W is tumor width (the longest tumor dimension perpendicular to L).
