Short term PD-1 stimulation alters monocyte levels and inhibits T cell proliferation. (A) Experimental setup. Ldlr−/− mice were fed a WTD or chow diet for 2 weeks while receiving an agonistic PD-1 antibody or control vehicle. (B) The weight and serum cholesterol levels were assessed before and after treatment. (C) Peripheral blood and (D) splenic monocyte percentages were measured by flow cytometry. (E) Proliferation of splenocytes after 3 days of stimulation with anti-CD3 and anti-CD28 measured by [3H]thymidine labeling (n = 5/group). (F) Regulatory B cells in peripheral blood were measured by flow cytometry. Data are displayed as mean ± SEM. Statistics was performed using one-way ANOVA, with post-hoc comparison using Dunnett's multiple comparisons test, comparing control chow and PD-1 WTD groups to the control WTD group.
