Model of current hypothesis. In control conditions, STAT3 is present at basal levels, and this acts as a brake on glutamatergic synaptogenesis. When the leptin receptor is activated in the presence of leptin, STAT3 activity is inhibited by the upregulation of KLF4 and SOCS3; this removes the brake and leads the formation of functional glutamatergic synapses in hippocampal neurons.
