A. DIAm fibers were classified based on immunoreactivity to anti-MyHCSlow, anti-MyHC2A, anti-MyHC2X, and anti-MyHCAll-but-2X. The photomicrograph shows representative immunoreactivity to anti-MyHCSlow and anti-MyHC2A in the same DIAm section using an IgG-Alexa-594 (red) and IgM-Cy5 (green) secondary antibody, respectively. Type IIx/IIb fibers were classified based on immunoreactivity to anti-MyHC2X and no or partial immunoreactivity to anti-MyHCAll-but-2X. Laminin immunoreactivity (blue) was used to help define muscle fiber sarcolemma. B. Cross-sectional areas varied across DIAm fiber types (p<0.001; one-way ANOVA). The cross-sectional areas of type I and IIa DIAm fibers were comparable and signficantly small than those of type IIx/IIb fibers (p<0.001; Tukey’s post hoc test).
