Increased MyD88-dependent IL-6 production and IL-17 expression and TH17 differentiation. Splenic B cells from MyD88-/- and MyD88+/+ mice were cultured either unstimulated or stimulated with LPS+IL-4 or anti-CD40+IL-4 in the presence of Zymosan, HKCA yeast or HKCA hyphae (A, B). After 5 days of culture, the concentration of secreted IL-10 (A) and IL-6 (B) was measured by ELISA. Data represent mean ± SEM of 4 mice per genotype, with triplicate measurements performed for each mouse. (C) Splenic B and T cells from wildtype mice were cocultured with anti-CD3ϵ and anti-IgM in the presence of Zymosan, HKCA hyphae or a TH17 differentiation inducing cocktail (anti-IL-4, anti-INF-γ, anti-IL-12, TGF-β1 and IL-6). The same culture conditions were also used in combination with a neutralizing IL-6 antibody. The percentage of IL-17+ cells among CD4+ T cells was measured by flow cytometry after 4 days of culture.
