(A, B) Double-immunostaining for myosin 7a and CtBP2 (presynaptic ribbon marker, white puncta) from the 45-kHz cochlear region of WT and Manf KO CBA mice show comparable numbers of ribbons at P56. (C) Ribbon numbers are quantified (CtBP2 puncta per inner hair cell [IHC]) in the 45-kHz-cochlear region. Quantification confirms a non-significant difference between the genotypes. P56: P-value 0.476 (linear mixed model, type 3 test of fixed effect, genotype); estimated marginal means WT 16.216 and KO 15.355; estimate of difference 0.861; WT 95% CI [14.036, 18.395]; KO 95% CI [13.458, 17.251].
