Quantitative suspension test using HPV16 pseudovirus. First, HPV16 pseudovirus (titer of at least 108 tissue culture infectious dose, 50% (TCID50/mL) and BSA (final concentration in the test procedure 0.3 g/L BSA) were mixed in equal volumes. This solution was then exposed to 8 volumes of the respective disinfectant (Table 1) and incubated at 20 °C ± 2 °C for the contact time indicated in Table 2. At the end of the contact time, cell culture media were added to the solution to stop the reaction. For determination of the virus titer after incubation with the respective disinfectants, 293TT cells (1 × 104 cells/well in 96-well plates) were incubated with 10−1 to 10−10 dilution series of HPV16 pseudovirus/disinfectant solution (n = 8 repeats for each dilution). Cells were analyzed after 48 h for GFP expression by fluorescence microscopy.
