To examine whether IP3 receptors were dysregulated in response to HIV infection, we determined mRNA, protein expression, and distribution in primary (indicated as astrocytes) and the U87 astrocytoma cell line transfected with CD4 and CCR5 (U87CD4CCR5). (A) IP3 receptor 1, 2, and 3 mRNA expression in primary and U87CD4CCR5 cells at 0, 3, 7- and 14-days post-infection. (B) Representative western blot for IP3 receptors 1, 2, and 3 in control (C) and HIV-conditions (H) at 3, 7, and 14 days post-infection. GAPDH was used as a loading control for the quantification shown in the panel. (C) Quantification of the IP3 receptor 1, 2, and 3 protein expression in primary and U87CD4CCR5 cells at 0, 3, 7- and 14-days post-infection.
