The effect of upregulation of miR-138 expression on the immune function and B cell differentiation of osteosarcoma Tfh cells. After Tfh cells were transfected with miR-138 mimics (miR-138) and its control mimic NC (mi-NC), (a) qRT-PCR was used to detect the expression of miR-138 in Tfh cells. (b–e) ELISA was used to detect IL-12 (b), IL-21 (c), INF-γ (d), and IL-10 (e) expression in the supernatant of Tfh cells. (f–h) Tfh cells were cocultured with B cells, and ELISA was used to detect the expression of IgM (f), IgG (g), and IgA (h) in the supernatant of each group of cells. (i) After Tfh cells were cocultured with B cells, the rate of CD27- and CD38-positive cells in B cells was detected by flow cytometry. ∗∗p < 0.01vs. HC group, ##p < 0.01vs. mi-NC/OS group.
