Phospho-substrate-trapping by the middle region of R15s. (a,b) R15 constructs were transfected into HEK 293T cells (input) and immunoprecipitated using anti-FLAG M2 magnetic beads (FLAG-IP). Samples were eluted from the beads by boiling in LDS and eluates were separated on a 4–12% Bis–Tris Plus gel. Proteins were detected by immunoblotting with FLAG, P-eIF2α, total eIF2α, PP1 and vinculin antibodies. Representative results of at least three experiments are shown.
