Strain YHE835 was transformed on dextrose media with either the vector (pH316) or the same plasmid containing HSP104 under the control of the GAL1 promoter (pA). Confirmed USA+ colonies of each were pooled and grown to single colonies on the indicated media. These single colonies were then tested for the ability to use USA for growth and for uracil feeding to a lawn of a ura2/ura2 strain.
