Figure 3. Vam6 and Vps3, are required for growth on heme and inorganic iron sources.

(A) The growth of vps3Δ, vam6Δ and vps3Δ vam6Δ double mutants and the WT strain was tested on yeast extract peptone dextrose (YPD), yeast nitrogen base (YNB) and YNB plus bathophenanthrolinedisulfonic acid (BPS) supplemented with either heme or FeCl₃ at 10 μM or 100 μM at pH 7.0. Tenfold serial dilutions of each strain (labeled on the right) were spotted on the indicated media after iron starvation and the plates were incubated at 30 °C for 2 days before being photographed. (B) Iron-starved cells of the indicated strains were inoculated in liquid YNB medium plus 150 μM BPS with and without supplementation with iron sources. A representative single mutant is shown for each gene and both double mutants are included. The cultures were incubated at 30°C, and OD₆₀₀ was measured. WT = wild type
