Effects of JAKi on JAK1/2 phosphorylation in IFN-γ stimulated neutrophils. Neutrophils were pretreated with JAKi (tofacitinib, baricitinib, upadacitinib) at the indicated concentrations for 1 h and then stimulated with IFN-γ (50 ng/ml) for 20 min. Phosphorylation of JAK1 (A) and JAK2 (B) were determined by Western blotting using phospho-specific antibodies against JAK1 and JAK2. Phosphorylation levels of JAK1 or JAK2 were normalized to total protein levels.
