Dexamethasone exerts its effects at the level of MAPK p38. HeLa-TO cells were transfected with 50 ng of pGL3c, 20 ng of pTetBBB-Cox2.5, and 100 ng of MKK6 expression vector (left panel) or constitutively active MAPKAPK-2 (MK2 Aspx3) expression vector (right panel). Dexamethasone (Dex) (1 μM) or SB203580 (SB) (1 μM) was added 22 h after the transfection, and tetracycline (Tet) was added after a further 2 h. RNase protection assays were performed as described in the legend to Fig. 2. (A) Representative experiment. Only the β-globin reporter and GAPDH loading control are shown.
