Scheme of platelets (PLT) preparation using density gradient centrifugation (DC). From the first centrifugation of whole blood with Ficoll-Paque^TM to the final pellet, which was resuspended in Dulbecco’s phosphate-buffered solution (DPBS) and ethylene glycol-bis (2-aminoethyl ether)-N,N,N′,N′-tetra-acetic acid (EGTA) at room temperature (RT). The PLT suspension was counted (10 times diluted) and added to the O2k-chamber at a final cell concentration of 200 × 10⁶ x·mL⁻¹. PBMC, peripheral blood mononuclear cell.
