UPR comprises three distinct signaling cascades initiated by IRE1α, PERK and ATF6 respectively, whose activation inhibits translation, boosts ER folding and degradation capacity, promotes ER biogenesis, and if the stress is unresolved, triggers cell death. The significance and molecular mechanism of the UPR in normal physiology and disease pathogenesis need to be further explored.
