a) Conditional Tsc1 knock-out (KO) in aortic smooth muscle cells led to significant experimentally measured changes in various species at gene and protein (intracellular and extracellular matrix) levels, as well as increases in cell proliferation, apoptosis, and degradative activity. The signaling network model qualitatively captured all of these experimental changes between the KO and baseline cells. b) Addition of the mTORC1 inhibitor rapamycin to the KO cells led to decreases in mTORC1-associated species and lysosome-related species, as well as an increase in contractile protein expression. The signaling network model also qualitatively captured these experimental observations for rapamycin administration in the KO mice. See also S2 Fig.
