In vivo transcriptional activation by selected amino acid substitutions in the VP16 core. A β-globin (OCTA+)TAATGARAT-containing reporter plasmid and an α-globin internal reference plasmid were transfected into HeLa cells in the absence (lane 1) or presence of wild-type (WT; lanes 2 and 12) or mutant (lanes 3 to 11) VP16 expression plasmids. RNA was collected 48 h after transfection and analyzed by RNase protection assay as described in Materials and Methods. The positions of correctly initiated α-globin (α), β-globin (β), and β-globin readthrough (RT) transcripts are indicated at the left.
