Mrgprb2 mediates the effects of lithocholic acid (LCA) in peritoneal mast cells (PMCs). (A) Mouse PMCs were treated with LCA (100 µM). An LCA-induced increase in intracellular calcium (n=2038 cells) was suppressed by pretreatment with 100 µM QWF in mouse PMCs (n=877 cells). (B) A comparison of relative responses of cells treated with LCA alone (left), LCA pretreated with 10 µM QWF (middle), or 100 µM QWF (right). (C) Representative fluorescent images of the PMCs at initial (0 s) and peak time points for 100 µM QWF pretreatment before LCA application. Scale bars=50 µm. (D) Results from β-hexosaminidase assay on PMCs treated with various LCA concentrations. Cp 48/80 indicates treatment of 100 µg/mL compound 48/80. **p<0.01 and ***p<0.001.
