At cellular level, WDR45 mutations cause autophagy impairment, alterations of ER morphology and quality control, perturbation of mitochondria morphology and function, and unbalances in iron homeostasis. These impairments result in protein aggregates accumulation, and enhanced oxidative, respiratory and ER stresses.
