Strain Δ70k lacks the soluble Tim9-Tim10 70-kDa complex. (A) Mitochondria from the parental (wild type [WT]) strain (10) and the Δ70k strain (Δ70k) were solubilized in 0.16% n-dodecylmaltoside and subjected to blue native gel electrophoresis (6 to 16% acrylamide) (27). Tim9 and Tim10 were detected by immunoblotting and incubation with ¹²⁵I-protein A. (B) Blue native gel electrophoresis was performed as in panel A; the blot was incubated with monospecific sera against Tim22 and Tim54. In the immunoblot with anti-Tim22p antibody (α-Tim22p), bands located above and below the 300-kDa band resulted from proteins that cross-react with the antisera (17).
