(A) Percentage of intact and defective HIV provirus detected in DNA samples from PBMCs of HIV-positive ART-treated individuals relative to all provirus quantified by IPDA. (B) Multiplex RT-ddPCR was performed to detect intact 5’ UTR containing transcripts (LTR) and 5’ UTR-deficient env transcripts (Env) using RNA samples prepared from PBMCs of HIV-positive ART-treated individuals. No RT enzyme control reactions were carried out for each assay and background signal was subtracted from experimental readings. Dots represent estimated copies/uL of single positive transcripts per μg of RNA. Dotted line represents the mean assay background signal calculated from five HIV-negative PBMC samples. Each point represents an independent participant sample. A total of 11 participant samples were assessed.
