miR-15a directly interacts with the 3′-UTR of GPX4. (A) The biotin RNA pull-down assay was performed to assess the enrichment of GPX4 following transfection with Bio-miR-15a-WT, Bio-miR-15a-MUT or Bio-NC. (B) Western blot analysis was performed to detect GPX4 protein expression following transfection with anti-miR-15a inhibitor. Luciferase activity of reporter plasmids containing (C) GPX4-UTR WT or (D) GPX4-UTR MUT miR-15a binding sequences of GPX4 3′-UTR, co-transfected with miR-15a mimics, anti-miR-15a inhibitor and the respective NCs.
