An illustration of our findings on spontaneous DNA double-strand breaks induction and their roles in maintaining their stemness and tumorigenicity of cancer cells. Leaky mitochondria lead to migration of EndoG and CAD from the mitochondria to the nucleus, which generates DNA DSBs and activation of ATM, which in turn activates NF-κB and Stat3 that confers stem-like properties to cancer cells.
