(A) IFN-β mRNA level in A549 cells infected by rgRSV mutants or rgRSV. A549 cells were infected by rgRSV-GALL(+), rgRSV-GALL(-), rgRSV-GALL(+/-) or rgRSV at an MOI of 0.5. At 18, 24, and 48 h post-infection, total RNA was harvested and IFN-β mRNA in cells was quantified by real-time RT-PCR. β-actin was used as an internal control. Error bars represent SD from n  =  3 independent experiments. (B) IFN-β mRNA level in A549 cells infected by rgRSV grown from METTL3-KD U2OS or WT U2OS cells with or without CHX treatment. A549 cells were treated for 1 h with 0 (UT, untreated) or 50 μg/ml CHX, and then infected either with rgRSV grown from METTL3-KD U2OS or WT U2OS cells at MOI of 0.5. At 24 h post-infection, total RNA was extracted and IFN-β mRNA was quantified by real-time RT-PCR. β-actin was used as an internal control.
