Figure S3. Effects of two kinds of RNA-aptamer on the spinal motor neurons of AR2 mice.

Figure S3.

(A, B) Number of ChAT-positive cells in the spinal anterior horns of the AR2 mice receiving either one of the aptamers at 20 μM or vehicle (n = 3 in each group) for 2 wk. Six anterior horns per one mouse were analyzed. The average number of cells in FN1040- or FN58-treated mice was significantly larger than in the vehicle-treated control mice and restored to the same level of wild-type mice. Two-way ANOVA followed by Tukey’s multiple-comparisons test. Mean ± SEM. *P < 0.05. (C, D) Size of the ChAT-positive cells expressed as mean diameter, two-way ANOVA followed by Tukey’s multiple-comparisons test. Mean ± SEM. ****P < 0.0001. (E) Proportion of ChAT-positive cells with nuclear TDP-43 (Nuc + Nuc/Cyto) was larger in FN1040- and FN58-treated mice than vehicle-treated mice. Total number of ChAT-positive cells used for TDP-43 localization analysis; C57BL6 (n = 230), vehicle (n = 118), FN1040 (n = 236), and FN58 (n = 210). Two-way ANOVA followed by Tukey’s multiple-comparisons test. Mean ± SEM. *P < 0.05, **P < 0.01 (Table S8, Blue bar; Nuc/Cyto).
