Figure S5. TNF selectively controls M2 gene transcription (related to Fig 4).

Figure S5.

(A) Identical samples like in Fig 4A and B were processed in the absence of reverse transcriptase and served as controls for genomic DNA contamination. (B) RNA from wild-type BMDMs left untreated (Ctrl), stimulated with IL4/IL13, IL4/IL13 + TNF, or TNF was used for qRT-PCR. TNF was added at the same time like IL4/IL13 (left), 12 h after IL4/IL13 stimulation (middle), or 18 h afterwards (right). The last time point was always 24 h of IL4/IL13 stimulation. qRT-PCR of Fn1 was analyzed. Data shown are the mean fold-increase of the Ctrl group. (C) Primary transcript qRT-PCR of the samples shown in (B). Superscripts show statistical significance between IL4/IL13 and IL4/IL13 + TNF. (D) Identical samples were processed in the absence of reverse transcriptase and served as controls for genomic DNA contamination. All values are means ± SEM; *P < 0.05; **P < 0.01; ***P < 0.001. Statistically significant differences were determined by two-way ANOVA. n = 3 biological replicates.
