IRE1α endoribonuclease domain inhibitor MKC-3946 decreased XBP1s, CHOP, ATF6 and ATF4 expression in vitro. (A-B) Western blotting shows the expression of XBP1s was induced by Ang II in a concentration and time dependent manner. (C) Western blotting shows IRE1α endoribonuclease domain inhibitor MKC-3946 can suppress the expression of XBP1s induced by Ang II in a concentration dependent manner. VSMCs were preincubated with MKC-3946 in various concentration for 30 min and then treated with Ang II (1.0 μM) for 12 hours. (D-E) Western blotting analysis of ER stress related protein in VSMCs. VSMCs were preincubated with MKC-3946 (40 μM) for 30 min and then treated with Ang II (1.0 μM) for 12 hours. MKC-3946 treatment can decrease XBP1s, CHOP, ATF6 and ATF4 expression.
