Simulations 11–20, wherein certain parameters (k1, k2, k3, k6, k7) shown in Figure 6 were varied. The units of k2, k3, k6, and k7 are min−1, and the units of k1 are nM−1min−1 [35]. The value of each of the 5 parameters for each of the 10 simulations are shown in the top part of the figure. The open horizontal bracket is the experimentally observed brain uptake of the HIRMAb-IDUA fusion protein in the Rhesus monkey, 1.2 ± 0.2%ID/100 g brain at 2 h after IV administration of an ID of 0.1 mg/kg [36].
