Determination of the effect of NS-398 on cell apoptosis and expression levels of cyclooxygenase-2, survivin, and prostaglandin E2 in human hepatocellular carcinoma (HCC) cells by flow cytometry and TUNEL assay. NS-398 (50 µM) and paeonol (Pae: 376 µM) were used to treat HCC cells for 24 hours [left (A,B,C,G,H,I): HepG2 cells; right (D,E,F,J,K,L): SMMC-7721 cells] separately. (A,B,C,D,E,F) Sub-G1 analysis by FACS. (G,H,I,J,K,L,M) Morphology and percentage of apoptotic cells by TUNEL staining. Cells with brown-stained nuclei were defined as apoptotic. [A,D,G,J: untreated cells; B,E,H,K: NS-398 (50 µM) treated cells; C,F,I,L: Pae (376 µM) treated cells]. Data are presented as mean ± standard deviation (SD) (**, P<0.01). FACS, fluorescence-activated cell sorting; TUNEL, terminal deoxynucleotidyl transferase dUTP nick end labeling.
