The effect of estrogen on interactions between EREs in BACE1 promoter and ERα, ERβ, REA, NcoR, and SRC-1 were detected by ChIP. Wild-type or ERα−/− mice received daily subcutaneous injections of E2 (20 µg) in 50 µl of 10% ethanol/sesame oil for 7 d; control animals received vehicle only. Mice were anesthetized, and ChIPs were performed using mouse brain samples. A, ERα interacts with ERE1, 2, 3, and 4 in WT mouse brains. B, ERβ does not interact with ERE1, 2, 3, and 4 in WT mouse brains. C, REA interacts with ERE1, 2, 3, and 4 in WT mouse brains. D, REA does not interact with ERE1, 2, 3, and 4 in ERα−/− mouse brains. E, NcoR does not interact with ERE1, 2, 3, and 4 in WT mouse brains. F, SRC-1 does not interact with ERE1, 2, 3, and 4 in WT mouse brains.
