Mechanism of action of glutamate and ivermectin on the GluCls at the nematode synapse. Glutamate is released from presynaptic neuron and binds to the extracellular domain of GluCl which leads to chloride ion influx, and hyperpolarization. Binding of ivermectin in the transmembrane channel domain of GluCl amplifies the effects of glutamate, allowing more influx of chloride ions, leading to hyperpolarization and paralysis of the invertebrate neuromuscular system.
