(A) Overnight cultures of the wild-type strain SC5314 and ire1Δ, mec1Δ, and vps15Δ mutants carrying a GFP-tagged FTR1 allele were inoculated in fresh YPD medium without or with BPS and grown for 5 h at 30°C. Cells were imaged by DIC and fluorescence microscopy. The two independently generated series of strains behaved identically and only one of them is shown. (B) Strains were grown as described above and analyzed by Western blotting with an anti-GFP antibody. Results for both independently generated series of strains are shown in each case.
