(A) Overnight cultures of the wild-type strain SC5314 and ire1Δ, mec1Δ, and vps15Δ mutants carrying a GFP-tagged MEP2 allele were inoculated in SD or SLAD medium and grown for 5 h at 30°C. Cells were imaged by DIC and fluorescence microscopy. The two independently generated series of strains behaved identically and only one of them is shown. (B) The wild type and ire1Δ mutants with GFP-tagged FTR1 or MEP2 alleles were grown in SLAD medium without iron and the cells observed as described above.
