The EC50 is defined as the concentration of stabilizer that affords 50% maximal activity (i.e., proteinase K protection) for fluorescein-labeled WIL-FL (WIL-FL*, 5 to 20 nM dimer) using the protease-coupled fluorescence polarization assay. The T46L/F49Y variant replaces two residues in the stabilizer binding site to the residues most commonly found in AL LC sequences34–35. Note that stabilizers 2 and 3 were referred to as 26 and (R)-83, respectively, in our previous study14.
