(A) HEK293 cells overexpressing FLAG-HA-tagged human Ago2 were lysed, and lysates were incubated with 200 nM of the indicated siRNAs for 4 h before immunoprecipitation of the Ago2 using an anti-HA antibody. Ago2-associated siRNAs were eluted by heating and quantified by stem-loop RT-qPCR. (B) Whole liver homogenates were prepared from C57BL/6 female mice (n = 3) treated with single subcutaneous injections of the either 2.5 mg/kg C5-targeted siRNA (sacrificed on day 5 post-dose) or 2 mg/kg Ttr-targeted siRNA (sacrificed on day 7 post-dose) Ago2-associated siRNAs were eluted by heating and quantified by stem-loop RT-qPCR.
