CK2 downregulation decreases the expression of JMJD2a and LSD1 at the translational level by activating the mTOR-p70S6K pathway. MCF-7 and HCT116 cells were transfected with (A) CK2α siRNA or pcDNA3.1-HA-CK2α, (B) CK2α siRNA in the presence of 50 μg/ml of cycloheximide (CHX) for the indicated times, or (C) CK2α siRNA or pcDNA3.1-HA-CK2α in the presence of 0.1 μM rapamycin. After 2 days, the level of each protein was determined via immunoblot analysis using antibodies against CK2α, hemagglutinin (HA, for HA-CK2α in Fig. 2A), JMJD2a, JMJD2c, LSD1, p-mTOR (S2448), and p70S6K (T389) (left panels). β-actin was used as a control. Graphs represent the quantification of each protein relative to β-actin (right panels). Data are shown as the mean ± SEM. *P < 0.05; **P < 0.01; ***P < 0.001. Bars that do not share a common letter (a-d) are significantly different among groups at P < 0.05.
