Establishment of organoid cultures generated from CRC patients (PDOs), subcutaneous xenografts (XDOs) or CTCs (CTCDOs). PDOs were directly generated from a colon adenocarcinoma surgically removed from a CRC patient. To obtain CTCDOs, PDOs were expanded, transduced with a luciferase-(LUC) encoding vector and orthotopically injected in the colon of immunocompromised mice. CTCs were then isolated from mouse peripheral blood collected by cardiac puncture and used to produce CTCDOs.
