Effects of α-T3E on proteasome activities in H2452 cells and H28 cells. (a) Cells were treated with 20 µM α-T3E for 0, 6, 12, or 24 h. After the treatment, the ubiquitinated protein levels in the samples were assessed by immunoblotting as described in Section 4.7. The α-Tubulin protein levels served as the loading control. The results are representative of two independent experiments. (b) After cells were treated with α-T3E (10–20 μM) for 24 h, chymotrypsin-like activity was assessed by a chemiluminescent method as described in Section 4.5.
