The orange box represents the conserved α-crystallin domain (ACD); the blue and green boxes represent the variable N-terminus and C-terminus domains (NTD, CTD, respectively); the red box indicates the I/V-X-I/V domains localizing at the NTD and/or CTD in several HSPBs; the purple box indicates the RLFDQxFG motif conserved in HSPBs. HSPBs functions include i) anti-aggregation activity against misfolded and aggregation-prone substrate and ii) cooperation with the degradative systems for substrate disposal, iii) protection against stressors and antiapoptotic activities, iv) cytoskeletal maintenance and dynamics.
