Galectin-3-mediated downstream processes such as CIE are altered by disruptions in O-GlcNAc cycling. HeLa cells were transfected with siRNA to knockdown either OGA (red) or OGT (blue). Clathrin Independent Endocytosis of CD59 was measured using a confocal imaging based antibody internalization assay, was found to increase when O-GlcNAc cycling was disrupted by either OGT or OGA knockdown (A), whereas clathrin mediated endocytosis of transferrin was not affected (B). At least three independent experiments were carried out with representative blots shown and data expressed as mean ± S.D.
