Identification of CgRIB1 encoding GCH2 in C. glabrata. (A) Yeast cells were cultured in SD medium until exponential phase. Serial dilutions of each strain were spotted onto SD medium with pH 5.5 in the presence or absence of RF and then further cultured. (B) The protein expressed from CAGL0F04279g in E. coli was purified. The purified protein was incubated in the reaction mixture over time, with or without pre-boiling. The resultant solution was reacted with diacetyl and then the fluorescence was measured. The fluorescence at time of 0 min was used as a blank in each sample. (C) Yeast cell-free extract prepared from cells cultured in SD medium for 24 h was used as a crude enzyme.
