Increases of collagen content and changes in organization in response to pulmonary artery banding (PAB) depend on sex and ovariectomy (OVX), but diastolic function does not. Effects of sex and OVX are shown for right ventricle (RV) interstitial collagen area (A), RV collagen organization into thick-to-thin collagen fibers (B), RV chamber compliance indexed to body weight (C), and tau Weiss (D). Representative images under brightfield (A) and polarized (B) light were captured at ×20 magnification. Collagen was stained with Picrosirius red; collagen area was calculated as the ratio of the area positive for collagen (stained in pink and red) to the total tissue area (stained yellow). Thick, tightly packed collagen and thin, loosely aligned collagen were identified as different interference colors under polarized light, with thick collagen defined as areas of yellow, orange, or red color and thin collagen defined as areas of green color. Note that OVX reduced RV chamber compliance index in Sham rats. n = 8–10 animals per group. Values are presented as means ± SE.
